Optimized dosing regimen of hydroxychloroquine for treatment of coronavirus disease 2019 using Monte Carlo simulation

Optimized dosage regimens of hydroxychloroquine (HCQ) in coronavirus disease 2019 (COVID-19) are currently unknown. We aimed to determine that rapidly achieved the pharmacokinetic-pharmacodynamic (PKPD) target for virological clearance COVID-19 patients. Plasma HCQ concentration was simulated using a non-steady state, 2-compartment linear model. The plasma trough (Ctrough) > 0.7 mg/L used as PKPD target. loading dose 800 mg three times daily and 1,200 twice on first day with probability attainment (PTA) 97.53% 82.63%, respectively. Maintenance 200 400 provided PTA 80% from 3 through 10 after initiation therapy. All proposed had < 1% achieve toxic level 4 mg/L. optimal early viral patients were followed by 9 days, days.
